We want to degrade mutant huntingtin protein causing Huntington's disease by proteasomal degradation and/or macro-autophagy. To achieve this, we would like to engineer ubiquitin ligases that selectively ubiquitylate huntingtin. We will then investigate with the engineered ubiquitin ligases how various types of ubiquitin chains change the fate of mutant huntingtin in mammalian cells.
